Defender Joleon Lescott (back) and midfielder James Milner (hamstring) failed to make the side at West Ham on Saturday and are unlikely to play.
Right-backs Micah Richards (knee) and Maicon (foot) and midfielders David Silva and Jack Rodwell (both hamstring) are again missing.
Defender Kolo Toure is not registered for the competition.
Ajax could select the same starting 11 that beat City 3-1 in the reverse fixture in Amsterdam last month.
Defender Niklas Moisander is available for selection again after missing the 2-0 league defeat to Vitesse at the weekend due to suspension.
City must defeat Ajax to retain hope of avoiding an early exit from the Champions League for the second successive season.
